Robert Hooke, 1660’s used an early compound microscope to view the structure of cork, leading to the term cell (cells in a prison).

Describe light microscopes

A

Light source passes through specimen, through lenses to reach viewer. Magnification up to 1500x. Resolution up to 0.2 micro metres. Specimen can be living or dead.

A

Sample labelled with fluorescent substance that attaches to the specific structures in the cell unable to be seen by light microscope. Sample illuminated with high-intensity light

Describe electron microscopes

A

Uses an electron beam, and electromagnets rather than lenses. Produces a computer image. Have a high resolving power (0.0002 micro metres). Magnification up to 1,500,000x. Specimen must be dead.
Two types:
Scanning EM - electrons reflected off specimen, 3D image
Transmission EM - electrons pass through, 2D

A

Confocal laser scanning microscopes use pinpoint laser beams to make images of many layers, which are combined into a 3D model.
