The Microscope Flashcards
What two structures are used to focus on the specimen?
Fine adjustment and coarse adjustment knob
Why do we never use the coarse adjustment knob on high power?
It can crack your slide
What happens if you use direct sunlight to illuminate your specimen?
You can damage your eyes
What are the objective lenses?
Low, medium and high power
What is the power of the low power objective lens?
4x
What is the power of the medium power lens?
10x
What is the power of the high power lens?
40x
What is the diameter of the field of view of low power?
4 mm
What is the diameter of the field of view under medium power?
1.5mm
What is the diameter of the field of view under high power?
0.4mm
How should you carry the microscope?
One hand gripping the arm, the other hand supporting the base
What are dry mounts for?
For objects that are not alive
What are the steps of a dry mount?
- place specimen on a dry slide
- cover with a cover slip
What are wet mounts for?
They permet the object to be better seen
What are the steps for a wet mount?
- place specimen on the slide
- with eyedropper, Place one drop of water on the specimen
- place cover slip at a 45° angle and let go
- blot excess water with a pencil eraser
How do you find the total magnification?
Ocular lens x objective lens magnification
How many micrometers are in one millimetre?
1000 micrometers in one millimetre
How many millimetres in 1000 micrometers?
1 millimetre in 1000 micrometers
How do you find the size of something under the microscope?
Diameter of the field of view
—————————————
Number of objects that fit across the diameter of field of view
What happened to the diameter of the field of you as you move from low to high magnification?
The diameter of the field of view gets smaller
Explain why the size of objects viewed under high-power is usually recorded in micrometres?
Millimetres are too large to measure the field of view. You need a smaller measurement which are micrometres
Which magnification would be the best for looking at several objects at the same time?
The low-power objective lens because it has the lowest magnification and the largest field of view so you can see all objects at the same time
