Techniques and Anesthesia Flashcards
What is the primary hematopoietic organ of the adult zebrafish?
Head kidney
Blood collection from fish under what size carries a high risk of death?
Less than 8cm
Describe ZF erythrocytes.
Nucleated, elliptical cells with eosinophilic cytoplasm
What is the most abundant granulocyte in ZF blood?
Heterophils.
Describe ZF thrombocytes.
Nucleated cells with dense nuclear chromatin.
Describe terminal and survival blood collection techniques in ZF.
Terminal: Decapitation through pelvic girdle, severing the heart. Dorsal aorta via transverse incision between the anal and caudal fins.
Survival: Dorsal aorta and posterior cardinal vein.
Survival blood collection in ZF has what mortality rate? How much blood can be retrieved?
2% mortality rate. Retrieve 2% of body weight.
What volume can be safely orally dosed to fish?
5 microliters
How does opercula movement correlate with anesthetic depth?
Decreased opercula movement with deeper anesthetic plane.
What are the advantages and disadvantages of anesthesia with Tricaine?
A: Moderate safety margin for short procedures, dose dependency, quick induction and recovery, and availability.
D: Acidic, requiring buffering, and may still be aversive when buffered.
What age of ZF has increased resistance to MS-222?
Larvae, compared to adults.
Can isoflurane be used as a sole anesthetic agent for ZF?
No, can be combined with MS-222
Describe use of lidocaine hydrochloride compared to MS-222.
Time to loss of equilibrium and recovery times is longer. May have a narrow safety margin (30% mortality).
What is eugenol also known as? What are advantages and disadvantages of its use?
Clove oil, Aqui-S, 4-allyl-2-methoxyphenol.
A: Low cost, availability, wider safety margin than MS-222, quick induction, and rapid metabolism and excretion.
D: Longer recovery times, potential for adverse reaction.
How is eugenol commonly prepared?
Mixed with ethanol, as it is not water soluble.
