Techniques Flashcards
1
Q
what are 6 types of contaminants
A
- Bacteria - usually introduced withe explant
- Fungi, yeast - enter on explant or air
- Insects - thrips, mites
- Initial contaminants - introduced with explant when disinfestation is insufficient
- Persistant or latent contamination - usually bacterial, show endogenously in plant tissue
- Introduced contamination - from poor technique
2
Q
Aseptic technique
A
without infection, avoiding contamination
- long hair tied back
- gloves, 70% ethanol
- no talking
- dont lean over work
- work in middle of hood
- dont disturb the air
3
Q
laminar flow hood
A
- removes particles from air
- air goes through HEPA filter
4
Q
surface sterilization
A
- alcohol (phytotoxic)
- Bleach (10-20 mins)
- Rinsing (sterile water 3X, 5 mins)
5
Q
explant
A
piece of plant that initiates tissue culture process
6
Q
callus
A
unorganized/undifferentiated mass of cells
7
Q
subculture
A
to remove a growing culture from a vessel to another
8
Q
accession
A
bring new plant material into culture
9
Q
sources of explants
A
immature embryos, cotylendonary petioles, shoots, roots, stems
10
Q
what can antibiotics be used for
A
- to cleanup contaminated tissue stocks or remove microorganisms used for transformation
- Agrobacterium tumefaciens - used to insert transgenic plasmids into plant cell
- timentin
