What type of differential stain is used for ones that don’t stain well?
Acid-fast stain
What type of differential stain is uses malachite green?
Endospore Stain
Stain procedure:
Crystal violet- cells stained purple
Iodine- acts as mordant, cells remain purple
Ethanol/acetone- decolorized, gram positive- purple, negative-colorless
Safranin- positive remain purple, negative turns pink
What does staining increase?
Contrast and resolution by coloring specimens w/ dye
What do microbiological stains contain?
Chromophore (colored portion of dye)
What are acidic dyes?
Acidic dyes stain alkaline structures
What are basic dyes?
(common) Stains acidic structures
What are special stains?
Negative (capsule) stain
Flagellar stain
