A

need around 50 SNPs to get the RMP of the core 13 STRs

Chip-based methods

A

DNA of known sequence attached in known place
DNA of unknown sequence with fluorescent probe added
Complementary then binds and fluoresces

TaqMan assay

A

add DNA probe with Q and R dye attached to PCR with special pol
if it binds then the pol breaks down probe and it amplifies and Q and R separate

Direct sequencing

A

PCR sample as usual
use ExoSAP to destroy remaining primers and dNTPs
PCR again with 1 primer and both dNTPs and ddNTPs
use electrophoresis to separate different sized pieces
look at fluorescence color to determine nucleotide

Minisequencing (SNaPshot)

A

same concept as direct sequencing but primers are designed to anneal directly before SNP and use only ddNTPs
