What is done after cleaning the slide?
Label the slide with the microbes you’re transferring. Make a dime sized circle on the underside of each slide.
What is done prior to obtaining bacteria from the liquid culture?
Flame your loop
Once you have a loopful of culture what do you do with it?
Spread the loopful of bacteria in the circle in the slide
How many loopfuls do you put on the slide?
2-3
What is done with the slide after the culture is added?
With a circular movement of the loop, spread the suspension into a thin area approximately the size of a dime
How much sample is needed from a slant (or plate)
Very little
What is added to the slide before bacteria when making a smear from a slant?
Water
Mix the culture and water to form what?
Milky emulsion
What is the next step after adding the sample to water and mixing?
Flame your loop

What does heat do?
Fixes slide which denatures enzymes. preserves size and shape cell when staining
How do you best fix the slide?
Quickly Pass slide through flame 2-3 times
