What amino acid is usually in the X and Y positions? Why?
Proline/ hydroxyproline so H-bonds can form because of the presence of the -OH group
What form of collagen is present BEFORE the protein enters the ER?
Pre-Procollagen (procollagen+Signal sequence to direct the protein to the ER)
How is Procollagen modified in the ER?
- Cleavage of signal peptide
- Hydroxylation and glycolation
- Disulphides bonds form between 3 alpha chains
- Triple helix forms between N and C terminals
What is scurvy a result of?
Weak tropocollagen triple helixs
How is a collagen fibril formed?
Tropocollogen is secreted out of the cell and the C and N terminal removed by proteases. Basic units line up and cross links form.
What is the name of the enzyme which form covalent cross links?
Lysyl Oxidase
What is Ehlers-Danlos Syndrome (EDS) as a result of?
Lysyl Oxidase deficiency
