Calculation of the cholesterol concentration
(Review)
Reaction principle
1 Serum cholesterol is determined after enzymatic hydrolysis and oxidation
Cholesterol ester + Water -> Cholesterol + Fatty Acid
2
Cholesterol + Oxygen -> cholestene-3-one + Water
3 The indicator quinoneimine is formed from hydrogen peroxide and 4-aminoantipyrine in the presence of phenol and peroxidase
2Hydrogen peroxide + 4-aminoantipyrine + phenol -> quinoneimine + 4Water
Specimen
Serum
EDTA- or heparinized plasma
Recommend a complete lipoprotein profile (TC, LDL-C, HDL-C, and TAG)
National Cholesterol Education Program (NCEP) Expert Panel on Detection, Evaluation, and Treatment of High Blood Cholesterol in Adults (Adult Treatment Panel III or ATP III)
Repeat testing at least once every ____
5 years
Reagent stability
1 Unopened - 2-8 C (expiry date)
2 Opened - 2-8 C (6 weeks) or 15-25 C (2 weeks)
