How do you culture a microorganism

1. Clean the bench with disinfectant solution . This killls microorganims to avoid contaminating the culture
2. Sterlise an inoculating loop by passing it over a bunsen burner
3. Open a sterile agar gel plate near a bunsen burner to kill all bacteria near the plate
4. Use the inoculating loop to spread the chosen bacteria all over the sterile plate
5. Close the agar plate with adhesive tape to prevent unwanted microorganism developing on the plate. Do not seal with tape all the way round as this will promote the growth of anearobic bacteria .place the agar plate upside down in an incubator to stop moisture dropping down and distrupting the microorgasims.Do this at 25 degrees to reduce the chances of harmful bacteria growing

How do you measure the effectiveness of antibiotics

4. Use the inoculating loop to spread the chosen bacteria all over the sterile plate
5. Place filter paper discs coated with different antibiotics onto the plate make sure you know which disc is coated with which antibiotic and make sure to leave space between each one . Have a filter paper coated in sterile water as a control so you know the growth in bacteria is down to the antibiotic only

7. Around the antibotics there will are zones where the bacteria dont grow called zones of inhibition
8. Measure the effect of teh antibiotic by comparing the size of the zones of inhibition . The larger the zone of inhibition the greater the antibiotics affectivness
