Research Methods Flashcards
Lesioning and microinjection
Electrolytic lesions or neurotoxin
Lesions can damage pathways
Microdialysis
Using cannula, collect NTs from specific brain areas and use HPLD to separate molecules and determine concentration
Electrophysiology
Stimulation and recording of neurons via electrodes, invented by neher and sakmann
Radio ligand binding
Measures quantity of receptors using gamma counter from homogenate
Must be:
1. Specific- measure with high affinity non radioactive ligand, measure remaining radioactivity (unbound)
2. Saturability
3. Reversibility and high affinity
4. Biological relevance-correlates with measurable effect in vivo
Receptor autoradiography
Location of receptors
Immunocytochemistry
Locate specific molecules within cells
In situ hybridization
Locate mRNA by using a fluorescent probe
In slice or homogenate
Construct validity
Extent to which test accurately measures characteristic being investigated
Face validity
Extent to which tests on animals can be generalized to humans
Predictive validity
extent to which lab test can predict clinical effects
Tail flick test
Latency to move tail when heat applied as measure of analgesia
Self administration test
Animals have access to a lever which they press to release the drug. Addiction measured by seeing e level a pain an animal will withstand to obtain the drug. Higher breaking point=larger reinforcement
Stereotactic surgery
Implantation of a device using stereotactic atlas based on bregma
