Protein Synthesis Flashcards
what is the central dogma of molecular biology?
DNA-RNA-Protein
What is a UTR
Untranslated region near the beginning of the mRNA, they are exons.
what does the 5β UTR do?
determines the rate at which the protein is synthesized
what does the 3β UTR do?
affects the stability of the RNA. (how long it can stay in the cell without being degraded)
How many possible codons are there?
64
How many types of ribosome are there?
2
one is bound to the ER
one is free in the cytosol
where in the ribosome does protein synthesis happen?
the cleft of the ribosome
What are the Ribosomal subunits?
60S subunit and 40S subunit which combined make up 80S ribosome
describe initiation
β cap binding protein associates with the cap and the poly A binding protein associates with the poly A tail.
βinitiation factors assemble on the small subunit and have GTP bound to it.
βa met-tRNA binds to the peptidyl binding site on the small ribosomal subunit.
βthe small subunit binds to the 5β cap of the mRNA and the ribosome moves along until it finds the AUG start codon which is next to a kozak sequence.
what happens once the start codon is found?
β60S binds to 40S forming complete 80S ribosome.
βEIF2 hydrolyzes the GTP to form GDP and releases pyrophosphate.
βAll EIFs are released from the ribosome.
What do viruses do that prevent antiviral proteins from being made?
inactivate EIF3 and EIF4
what is the enzyme that moves the amino acid in the P site to the A site?
peptidyl transferase
describe elongation
β tRNA with the next amino acid comes to the A site which involves elongation factor 1
βwhen the covalent bond forms between methionine and the next amino acid, peptidyl transferase moves the amino acid in the P site to the A site
β the tRNA on the P site is now empty
βthe tRNA in the A site has both amino acids attached to it.
βP site tRNA now moves to the E site so it can go and get recharged so a new tRNA can enter the A site.
what is translocation and what does it require?
βtRNAs moving from the A & P sites to the P & E sites respectively.
βuses up one GTP
β the enzyme translocase and EF2.
what enzyme recharges tRNA?
βaminoacyl tRNA synthase
