Polymerase Chain reaction Flashcards
What is the main role of the PCR?
Polymerase Chain reaction is used to make multiple copies of specific sequence of DNA called the target DNA.
What is the method it uses ?
Heating and cooling methods to make many copies of a specific region of DNA
What is the first step ?
First the temp is raised to near boiling causing the double strands DNA to seperate or denature in to single strands
When the temp is decreased, short DNA sequence known as primers bind to complementary matches on the target DNA sequence
Primers then bracket the target DNA sequence to be copied
Enzymes the DNA polymerase forms new DNA strands sequences ,adds nucleotides to extend second strands this completes the first cycle
Cycle 2
The process is repeated ,denaturing , annealing are repeated
What occurs after three cycles?
The target sequence defined by the primers begin to accumulate
After 30 cycles
A billion copies of the target sequence are produced from a single starting molecule
