factors associated with serious ADR
A
PK is altered, steep dose response or adverse response relationship, toxicity within or near therapeutic range, absorb/elim via a single rate-limited pathway, low oral availability/high first-pass metabolism (a change = huge increase in dose), unusual PK , especially zero order or saturation (concentration takes off once you have reached the saturation is met) - small increments in dose with these drugs
