pcr and reverse transcriptase Flashcards
what are the three main stages in the use of pcr
denaturation , annealing, dna synthasis
what is the purpouse of pcr
used to amplify dna sample in vitro = allows losts of dna to be formed from a small sample
what is a limitation of pcr
can only use a sample of dna max 1000
define and explain the process first stage of pcr
denaturing where the template strand to be copied is seperated to a singe strand- this is done by placing the strand in a thermocycler and heated to 95 degrees inorder to destablise and break the hydrogen bonds forming a single strand
define and explain the process in the second stage of pcr
annealing - cooled to 65 degrees
- primer added
- primers anneal to strands
what is the third stage of pcr and explain the process
dna synthesis -
- solution heated to75 degrees
- dna polymerase binds complementory and free nucleotides
how many strands of dna does one cycle of pcr produce
2
what is the formular to work out no. of pcr produceed
n=2x ( where x is no. of cycles and n is no. of dna molecules
what is the differance between a sense strand and an anti-sense strand.
sense strands gode for specific protiens
antisense strands are non coding
