acetaminophen
mechanism
-overdose= overwhelmed liver
-NAPQI (N-acetyl-p-benzoquinomimined toxin released
-usually liver neutralized NAPQI with glutathione
-when overdosed glutathione store becomes depleated
-NAPQI builds up and causes oxidatitive stress to liver cells
liver damage
NAPQI accumulates and attaches to hepatocytes and causes cells disruption
-the liver cant filter other substances so toxins build up
symptoms
nausea, vomiting, general malaise
-jaundice,abdomial pain, confusion , swelling
-could lead to liver failure, bleeding and multiorgan failure
treatment
-NAC (n-acetylcysteein) which replenashes gluthione levels and neutralises NAPQI
If caught every enough liver can fully recover
-3 doses over 21 hours IV
