Page 2 Flashcards
Legal’s Test steps
3 cc urine + NaOH or KOH -> alkaline urine
Few drops Acetic acid (concentrated)
Few drops Na nitroprusside, red to purple if (+)
Legal’s test detects what ketone body exactly?
Acetone
Acetoacetic acid
Rothera’s Test steps
5 cc urine (fresh) + 1 gm of NH4 SO3
2 drops Na nitroprusside
NaOH overlay, yields purple color when (+)
Negative when no purple color after 10-15 minutes.
Rothera’s Test is negative, result?
No rxn after 10-15 minutes
Acetest steps
Put Acetest reagent tablet on surface
Few drops urine on tablet, turns purple after 30s if (+), compare to color chart
Which ketone tests uses a color chart?
Reagent Strip Test
Acetest
Manner of Reporting acetone for acetest
+1 (Trace)
: 25-50 mg%
+2 (Moderate)
: 200-250 mg%
+3 (Strong)
: 400-1000 mg%
Manner of Reporting diacetic acid for acetest
+1 (Trace)
: 10-20 mg%
+2 (Moderate)
: 25-40 mg%
+3 (Strong)
: >50 mg%
Why KOH or NaOH is added in Legal’’s Test
To produce an alkaline urine
