P53 Flashcards
What are some P53 therapeutics?
- Gendicine
- Onyx adenovirus (oncorine)
- Nutlins
What does gendicine do and what cells do they attack? what is the before treatment or after treatment?
P53 negative cells
they replace the P53 gene by using an adenovirus to deliever P53
before treatment: tumor has +P53 and -P53
after treatment: all tumor cells have +P53
what does onyx adenovirus (oncorine) and what cells do they target? what is the before treatment or after treatment?
they target P53 negative cells
it is a lytic virus (lysing and spreading through tumor cells)
only replicating in/infecting cells that are lacking P53 function
before treatment: tumor has +P53 and -P53
after treatment: tumor has only +P53 and the cells were lysed
what does nutlins and what cells do they target? what is the before treatment or after treatment?
they target +P53 cells
small molecule protien modifier
need to have P53 produced in cell for this therapeutic to be effective
sequesters MDM2 to increase P53 activity
before treatment: tumor has +P53 and -P53
after treatment: P53 is activated
What does HPV and cervical cancer do? what happens with E7 and E6?
E6: binds to p53 and inhibits its function
E7: binds to the RB protein and prevents the interaction of E2F
Fill the blank: ___ of cancer have mutated P53
50%
how does P53 regulate MDM2 protein?
MDM2 binds to N-terminal P53 transactivation domain; resulting in
no phosphorylation/activation of P53 and translocation of P53 into cytoplasm
