Organoids and Organs on a Chip Flashcards
What is an Organoid
small and self organized, 3D tissue cultures or cell clusters derived mainly from adult stem cells
What is an Organoids function?
Organoids serve as powerful models for studying organ development, disease modeling, drug screening, and regenerative medicine. They provide a platform for investigating cellular behaviors, interactions, and responses in a more physiologically relevant context compared to traditional two-dimensional cell cultures.
What are properties of organoids?
Organ specific
Acts similarly to the function of the organ
Spatially organized similar to the organ
Types of 3D cell cultures
scaffold and scaffold free
Scaffold 3D cell culture
-solid scaffolds like hydrogels
-mimics ECM
Scaffold free 3D cell culture
-independent of scaffolds
- centrifuging nonadherent coatings
Scaffold 3D cell culture
use polymers such as polystyrene. 3D culture is made by growing cells on a prefabricated scaffolds of polymers that mimic ECM.
What is Organ on a Chip
Micro-engineered biomimetic system. Engineered microchips containing living human cells that reconstitute organ level function.
Why is organ on a chip useful
-to test efficiency, toxicity of drugs and chemicals
-to create in vitro models of human disease
RE> 4000
turbulent, unpredictable
2300<RE<4000
transitional, turbulent outburts
RE<2300
Laminar, predictable slow mixing
Fabrication methods
Soft lithography-used PDMS
Photolithography- uses UV light
