in all normal tissue- produces prostaglandins that are protective to gastric mucosa- pgi2, pge2
cox 2 is found
at site of inflammation, not normal
nsaids do what
inhibit cox2 (pain, inflammation, fever), but give unwanted cox1 results (gi ulceration)
arachoidonic acid comes from where
cell membrane, cleaved off by phopholipase a2
2 pathways arachoidonic acid can taeke
lipogenase or cyclogenase
lox pathway
arachoidonic acid is turned into leukotrienes by lipogenase
product of lox pathway
lta4–>ltb4—>ltc4—>ltd4. these bind to leukotriene receptor and cause bronchoconstriction
cox pathway
arachidonic acid is converted to prostaglandin h2 by cyclogenase (COX)
3 products of prostaglandin H2
in platelet creates thromboxane A2 which is bad for the heart
2. in the endothelium makes prostacyclin (PGI2) which is good for the heart
3. turns into prostaglandins which are good or bad-
pge2 and pgf2 are good for the stomach (protective) COX1
pgd2 causes inflammation, fever and pain (COX2)
