Name some general properties that NSAIDs have in common.
A
- All but one are weak organic acids (nabumetone, a ketone prodrug that is metabolized to the acidic active drug)
- most are well absorbed, food does not sign. change their bioavailability
- highly metabolized (largely by liver, cyp3A or cyp2c families of P450 enzymes)
- Renal excretion is the most important final route, but nearly all undergo some degree of bilirary excretion & reabsorption
- most highly protein bound, usually to albumin
- most are racemic mixtures
- all can be found in synovial fluid after repeated dosing
