Non-Cell Cycle Specific Flashcards
Alkylation agents
Cytotoxic effects related to alkylation of reactive amines, oxygens, or phosphates on DNA
Acute effects manifests against rapidly proliferating tissues
AE: carcinogenic in nature (inc risk of 2 malignancies), vesicants, toxic effects on make and female repro
Nitrogen mustards
Cyclophosphamide, Ifosfamide Beware of water intoxication from adequate hydration Urothelial damage Severe hemorrhagic cystitis Metabolized by CYP450 Mesna conjugated acrolein in urine
Busulfan
Alkyl sulfonates
Causes pulmonary fibrosis
Leukocyte count may cont to drop for a month after discontinuation
Platinum analogs
Cisplatin, Carboplatin
Oto and nephrotoxic
Not cross resistant with oxaliplatin
Doxorubicin
Abx
Irreversible cardiomyopathy
Generates free radicals
Metabolized in the liver
Cardiotoxicity:
Acute- two to three days arrythmias occur and conduction abnormalities
Chronic- dose dependent dilated cardiomyopathy associated with heart failure
