Q

Mécanismes physiopathologiques de la protéine tau

A

Différentes isoformes de tau (3R-tau, or 4R-tau, or mixed 3R-/4R-tau isoforms),

Génération d’agrégat de différentes formes :
=> paired helical
=> straight filaments)

Atteinte de différents types cellulaires
=> astrocytes
=> oligodendrocytes
=> neurons

Différents morphologies des agrégats
=> spherical Pick’ bodies vs neurofilamentous tangles in neurons
=> tufted astrocytes vs. astrocytic plaques

Atteinte de différentes régions du cerveau.

Q

Types de tauopathie ?

A

Primary tauopathie

4R tauopathie :
=> Corticobasal degeneration (CBD)
=> Progressive supranuclear palsy (PSP)
=> Globular glial tauopathy tauopathy (GGT)
=> Argyrophilic grain disease (AGD)

3R tauopathie
=> Maladie de Pick

Mix 3R/4R tauopathie : NFT demencia (, characterized by NFT in the area of the medial temporal cortex and ammons horn without associated beta-amyloid deposits.

Secondary tauopathie
(including Alzheimer’s disease, where tau pathology occurs as a consequence of amyloid-beta pathology)
