What is LGX?
A
LGX is a more rapid, detection tool of MRSA and MSSA which should reduce the spread, reduce length of hospital stays and associated treatments costs.

Issues with PCR/Cell culture
A
Requires lab and trained technicians
Can take up to 22 hours for PCR and 65 hours for cell culture

LGX mechanism
A
Stephylocoagulase enzyme forms a complex with prothrombin and cleaves are specific set of amino acids - valine, arginine and proline. LGX is colourless prior to cleaving and coloured once cleaved.

Rhodamine two forms
A
- Cyclic ester group with electron density being pulled away by amines on either side. (colourless)
- Zwitterion form (coloured)

LGX testing
A
Tested on live bacteria
- Flourescence response is immediate with SA
- E.coli has a very slow response
- CNS has a much lower than negative response as they form clumps and diffract light

LGX and prothrombin are mixed in buffer when cylinder is pushed down
