Monoamine oxidase inhibitors Flashcards
Why are serotonin, norepinephrine and dopamine are called mono-amines ?
They only have one amine group.
What is the action of norepinephrine in the CNS ?
It is responsible for enhancing alertness and focus.
What are the main actions of dopamine?
It modulates cognitive function, motivation, and awakens.
What is the action of monoamine oxidase A ?
The presynaptic degradation of serotonin, norepinephrine and dopamine.
What is the action of monoamine oxidase B?
It only breaks down dopamine.
MAOIs are used as what line of agent for the Tx of MDD?
Second or third line.
What is the first line Tx in atypical depression ?
MAOIs.
What are the non-selective MAOIs and their MOA ?
Isocarboxazid, Phenelzine, and Tranylcypromine. They irreversibly inhibit both MOAI-A and B.
What are the selective MAOIs and their MOA ?
These are selective inhibitors of MAOI-B responsible for dopamine degradation and they are Seligilin and Rasagilin namely.
What are the most dangerous Side effects of MAOIs?
Serotonin syndrome and hypertensive crisis.
What is the trigger of hypertensive crisis in patients treated with MAOI ?
It is triggered by the consumption of Thyramine rich food such as cheese, beer etc while taking MAOIs. The inhibition of thyramine breakdown due to the inhibition of MAOIs in GIT leads to increased circulating Thyramine which in turn increase the release of norepinephrine leading to hypertensive crisis.
What is the Tx of MAOIs induced hypertensive crisis ?
Adrenergic antagonist Phentolamine.
Selegiline does not present with a risk of hypertensive crisis as it selectively inhibits_______
MAOI-B
________are the two classes of antidepressants that make up the 1st generation antidepressants.
TCAs and MAOIs.
__________are MAOIs that are only given orally.
Phenelzine and tranylcypromine
