Microscopy Flashcards
What does resolution mean?
How well a microscope distinguishes between two points that a close together
What are the differences between a light and electron microscope?
Electron microscope can’t view live cells, while light microscopes can
Electron microscopes have higher resolution and magnification, which means we can see smaller things in much more detail
Name the 8 steps to view a specimen using a light microscope
1.) Take a thin slice of the specimen to allow light to pass through it
2.) Take a slide and place the specimen onto it
3.) Add a drop of stain and make it easier to see
4.) Place a cover slip at a 45⁰ angle to prevent air bubbles and clip the slide onto the stage
5.) Select the lowest- powered objective lens
6.) Use the coarse adjustment knob to move the stage up and down until it is nearly in focus.
7.) Then adjust the fine adjustment knob until you have a clear image
8.) Get a clear ruler and measure your field of view
How do you create a scientific drawing of a specimen?
1.) With a sharp pencil, draw outlines of the main features with clear, unbroken lines and no colour
2.) Keep all parts in proportion
3.) Label important features and include the magnification with a scale
Total magnification = ?
Objective lens × eyepiece lens
What is the formula for magnification?
Image size ÷ actual size
