microscopy Flashcards
1
Q
what is a microscope ?
A
piece of equipment that allows us to view small specimen such as cells that cannot be seen with the naked eye
2
Q
what are the two types of microscopes ?
A
electron microscope
light microscope
3
Q
light microscope
A
- lower magnification, lower resolution
- coloured image
- can be used on dead and alive cells
- easier to use, lightweight
- uses light rays
4
Q
electron microscope
A
- uses beams of electrons
- higher magnification and higher resolution
- only black images
- only uses on dead cells
- much harder to use, requires special training
5
Q
how to view specimens with a light microscope
A
- take a thin slice
- put a drop of water in the middle of slide
- carefully put specimen over the slide
- stain it with iodine
- use mounted needle to put coverslip
- place slide on the stage, put stage clips over to secure the slide
6
Q
how to view specimens with a light microscope p2
A
- select lowest powered objective lens
- use rough focus to move stage up, look down eyepiece and move stage till specimen is in focus
- use fine focus to adjust till you get a clear image
7
Q
how to calculate magnification
A
image size / actual size
8
Q
second magnification formula
A
eye piece lens * objective lens power
9
Q
millimetre
A
10 to the power -3
