Microscopes Flashcards
The electron microscope
The electron microscope is a type of microscope that uses a beam of electrons to create an image of the specimen. It is capable of much higher magnifications and has a greater resolving power than a light microscope, allowing it to see much smaller objects in finer detail.
what are the limitations of using an electron microscope?
Specimen preparation is crucial and you can’t view live organisms as it has to be in a vacuum
What are the common staining agents of TEM?
Uranium and Lead
What does SEM produce a picture of?
Cell surface
What is FRAP used for?
determine the kinetics of protein redistribution/protein trafficking and to detect how much protein is immobile and how much is mobile
What are the disadvantages of using GFP?
Photobleaching Cellular distribution Protein folding Over expression Phytotoxicity
