microscope Flashcards
A tool used in magnifying and illuminating specimens that are otherwise invisible to the naked eye
microscope
when was microscope developed
1590
who invented the microscope
hans and zacharias jansen
he described the microscopic appearance of cork and used the term cell to describe the compartments he observed (give the year too)
1667, robert hooke
observe living cells under the
microscope, which he named “animalcules” (give year too)
1675, anton von leewenhoek
identified the cell nucleus
1830, robert brown
who gave rise to the cell theory
matthias schleiden, theodor schwann
2 main parts of microscope
mechanical and optical parts
optical parts
eyepiece, mirror, objective lens
3 types of obj lens and its magnificaiton
Oil immersion objective - 100x
High power objective - 40x
Low power objective - 10x
types of microscope
Stereo microscope Compound microscope
Inverted microscope Metallurgical microscope
Polarizing microscope
Optical microscope Electron microscope Scanning probe microscope
types of electron microscope
Transmission electron microscope (TEM) Scanning electron microscope (SEM)
types of scanning probe microscopes
atomic force microscope
scanning near-field optical microscope
simplest optical microscope
magnifying glass
the lens at the top that you look through
eyepiece lens
magnification power of eyepiece lens
10x
connects the eyepiece to the objective lenses
tube
supports the tube and connects it to the base. it is used along with the base to carry the microscope
arm
the bottom of the microscope used for support
base
a steady light source used in place of mirror
illuminator
the flat platform where you place your slides. stage clips hold the slides in place
stage
this is the part that holds 2 or more objective lenses and can be rotated to easily change power
revolving nosepiece
this is an adjustment that determines how close the objective lens can get to the slide
rack stop
this is used to focus the microscope. it is always used first and it is used only with the low power objective
coarse adjustment
this is used to focus the microscope. it is used with the high power objective to bring the specimen into bette rfocus
fine adjustment
microscope care and handling
Transporting
⚬ grab the arm with one hand and place your other
hand on the bottom of the base * Handling
⚬ never touch the lens with your fingers * Cleaning
⚬ only use lens paper to clean the glass * Storage
⚬ put the low power objective in place and the stage all the way up
used to vary the intensity and size of the cone of light projected upward into the slide
diaphragm/iris
