MANUAL: PART 3: Medium and materials Flashcards
It contains suitable environment and nutrients for microbes to grow.
Culture medium
A solidifying agent extracted from seaweed composed of complex sugars: liquid at 100° C and solid at 40°C.
Agar
A medium to observed morphologies of microorganism. Generally requires 15-20ml of molten agar.
Agar plate
A medium that generally requires 7ml of molted agar dispensed into tubes before autoclaving and slanted after sterilization.
Agar slant
A medium where no agar or solidifying agent is used.
Broth
A subculturing instrument where it is used to transfer small amount of microbes to plates, tubes, and broth.
Inoculating loop
A subculturing instrument where it is used to transfer microbes to broth by stabbing into agar deep tube or semisolid media.
Inoculating needle
A subculturing instrument, it is used to transfer culture media from one vessel to another.
Pipette
A type of pipette where it has etched ring on mouthpiece.
Serological pipette
A type of pipette where it has no etched ring.
Mohr pipette
An equipment used for destroying all forms of life: set at 15 minutes; 15 psi; and 121°C.
Autoclave
An equipment use to maintain optimum temperature for microorganism to grow: typically set at 37°C, for 18-24 hours.
Incubator
An equipment use to storage and make microbes bacteriostatic. Typically set at 4°C.
