Levodopa

A

(= L-DOPA, Dihydroxy-Phenylalanine)
- Parkinson’s disease

MOA: prodrug, wird im Hirn zu Dopamin
wird auch Tyrosine gebildet
+ Decarboxylasehemmer, damit in Peripherie gehemmt
Food-Drug Interaction:
1. competing with amino acids at jejunal transporters
2. Amino acids inhibit levodopa transport via blood-brain-barrier → less levodopa in the brain

→ levodopa should be given 60 mins prior to meals, do not put in yoghurt (auch wenn dysphagia in PD oft vorkommt)

Hypericum / Hyperforin / Hypericin

A

Hypericum perforatum

MOA: reuptake inhibition of serotonin and norepinephrine → antidepressant effect

  • Induction of CYP 3A4 & CYP 1A2 (nicht Hemmung!)
    e. g. transplant rejection because self-medication with St. Johns wort and thus inefficient Immunosuppressant (Cyclosporin, wurde durch Induction sofort abgebaut)
