Lecture 8- How are Proteins Studied Outside Cells Flashcards
Test of Discovery
try to find out the location of our protein of interest
Experimental group
we do not know what’s going to happen
Positive control group
event should occur
Negative control group
even shouldn’t occur
Technical controls
done to quantify the results of experimental and control groups
Purpose of SDS-PAGE
to separate proteins by size
Steps in SDS-PAGE
- coat all the proteins with negative molecules (SDS)
- load gel (hinder their movement)
- Run gel
- soak gel in coomassie blue
Coomassie blue staining
Purpose: to make all the proteins blue
Molecular weight markers
tells us the size of our band
also called MW ladders, protein standards
Western Blotting
shows a specific protein on a gel
Detection reagents
colorless, but the enzyme can react and turn them into a blue precipitate.
Loading control
antibodies against a protein that should be present in all samples
