Lecture 28 Flashcards
James watson
american chemist
Francis Crick
english molecular biologist
Rosalind franklin
British chemist and microphotographer
Maurice Wilkins
british researcher; discovered how to prepare uniformly oriented DNA fibers
Linus Pauling
was completely wrong about the structure of DNA
xray crystallography
x ray diffraction; crystals of a molecule are bombarded with a beam of xrays; the rays are bent (diffracted) by the molecules and the pattern is recorded on photographic film
Erwin Chargaff
nucleotide composition of DNA molecules vary in complex ways suggested that DNA was not a simple repeating polymer; discovered that ratio of A= ratio of T and ratio of C = ratio of G
Ball and stick models
watson and crick onoly built models and were viewed as lazy; watson went to franklin results report lecture as a spy and built a model but forgot the water content
X pattern
Franklin discovered the helix through the picture coming out looking like an X, but wilkins shows watson his picture; watson figured out helix based on that
nucleotide
3 main components: 5 carbon sugar, phosphate group (makes it an acid), and nitrogenous base
Adenine, Guanine
purine bases
Cytosine, thymine, uracil
pyrimidine bases
double helix
very stable with spacing of 3.4 angstroms
phosphodiester backbone
composed of alternating sugar and phosphate groups
