Lecture #2-1 Flashcards
Name some cytokines that regulate hematopoesis.
Stem Cell FactorIL-1, IL-3, IL-6, IL-7EPOColony Stimulating Factors
What cells come from a hemocytoblast?
All blood cells. Immediately, common lymphoid and myeloid progenitors.
What comes from the common lymphoid progenitor cell
Small lymphocytes that develop into B cells, T cells, or NK cells.
What comes from common myeloid progenitor cells
Megakaryocytes, erythrocytes, and myeloblasts.
What four cells types come from myeloblasts?
Basophil, Neutrophil, Eosinophil, Monocyte
Three sites of hematopoesis over the course of development.
Yolk sac, fetal liver/spleen, bone marrow.
Name the two traits that hematopoetic stem cells are known for.
- Self Renewal2. Totipotency
Where can adult hematopoetic stem cells be found?
Bone Marrow
Where do T cell progenitors mature? B cell?
Thymus, Bone Marrow
Describe the nucleus of a neutrophil.
Tri-lobed
Describe the nucleus of a eosinophil.
Bi-lobed
Describe a basophil cell
So full of blue granules its hard to see anything.
Monocyte? Macrophage?
Pac Man, Pac man with vacuoles.
Describe the appearance of a megakaryocyte.
Big and amorphous…maybe a bit jagged.
Describe the appearance of a NK cell, lymphocyte.
Lymphocyte – Round nucleus that takes basically all of the cell. NK cell – Rounded like a lymphocyte, but slightly more amorphous and smaller nucleus by comparison.
Neutrophils…name the origin, antigen receptor, function, marker, and induction factor.
Bone MarrowNonePhagocytosis/Killing MicroorganismsCD15G-CSF
Monocytes…name the origin, antigen receptor, function, marker, and induction factor.
Bone MarrowNoneBecome Macrophages/DCsCD14M-CSF
Eosinophils…name the origin, antigen receptor, function, marker, and induction factor.
Bone MarrowNoneKilling Ab coated parasites via cytotoxic granulesIgE and IgG ReceptorsIL-5
Mast Cells…name the origin, antigen receptor, function, marker, and induction factor.
Bone MarrowNoneTo Kill Parasites (Allergies as a bonus), make IL-4/5/6/13IgESCF
Basophils…name the origin, antigen receptor, function, and marker.
Bone MarrowNoneEffector cells in allergic disorders and parasite defense.IgE receptor
NK Cells…name the origin, antigen receptor, function, marker.
Bone Marrow and ThymusNoneKill tumor+virus infected cellsHas activating and inhibitory receptorsCD16, CD 56
T cells…name the origin, antigen receptor, function, marker.
BM, Thymus, Differentiation in LNYesRegulate Humoral and Cellular Immune ResponsesCD3 (all Ts), CD4, CD8
B Cells…name the origin, antigen receptor, function, marker.
Bone MarrowYesB Cell ReceptorProduction of Ab (GAME)CD19/CD20
What cells can B lymphocytes mature into? What is required for this to happen?
B Plasma Cells with the help of Antigens and T cells
Describe the Portions of WBC of each cell type.
Neutrophil – 40-75%Lymphocyte – 20-50%Monocyte – 2-10%Eosinophil – 1-6%Basophil <1%
How are hematopoetic stem cells used in therapy?
Bone Marrow Transplant for immunodeficient/cancer
What can you give a person to help boost neutrophils and monocytes by increasing leukocyte production?
G-CSF or GM-CSF
How to medically induce RBC creation?
EPO
NK Effector machinery?
Perforins and Granzymes
