Lecture 14 Flashcards
1
Q
How are restriction enzymes prevented from cutting DNA?
A
Adenine is methylated.
2
Q
What is the difference between dideoxy DNA and normal DNA?
A
ddDNA doesn’t have a hydroxyl group that can be added to so it terminates the chain.
3
Q
What are the steps to Southern Blotting?
A
1) DNA is cleaved with restriction enzymes.
2) DNA fragments are separated by size using gel electrophoresis
3) DNA fragments are then transferred from gel to nitrocellulose using the capillary action induced by a alkaline solution.
4) Hybridize fragment of interest with labeled DNA or RNA probe.
4
Q
What is the subject of Northern Blotting?
A
RNA
5
Q
What is the subject of Western Blotting?
A
Proteins
