Lecture 11 Flashcards
What are the main Enzymes for DNA Replication?
Helicase, DNA polymerase I, SSB, Primase, DNA polymerase III and Ligase
What did Griffiths do?
Discovered there was a transforming principle by infecting mice with non-virulent and virulent disease
What did Avery, MacLeod and McCarty do?
Discovered that DNA is the transforming principle by repeating Griffiths experiment but using enzymes to degrade certain macromolecules
What did Hershey and Chase do?
Discovered DNA was the universal transforming principle by using radioactivity and a blender to test proteins and DNA
What did Watson and Crick do?
Discovered DNA is a double Helix
What did Meselson-Stahl do?
Discovered that DNA replicates semi-conservatively by testing weights of Nitrogen in DNA
What does Helicase do?
Unzips DNA
What does SSB do?
Single Strand Binding proteins bind tot he Single Strands to keep them from closing into Double Strands
What does Primase do?
Adds an RNA primer to the 3’ end but the primer is 5’
What does DNA polymerase III do ?
Extends off primer, asking DNA 5’ to 3’
What does DNA polymerase I do?
Removes primer with DNA
What does ligase do?
Ligase joins DNA fragments together
