principle:
addition of glass beads to break neutrophil to expose the nucleus to be engulfed the macrophage

- Incubate tube of blood at 37C for 30 minutes
- Fill 3 to 4 Wintrobe tubes with the well mixed blood and centrifuge at 200g for 10 minutes
- Discard the plasma from the tubes. Remove the buffy coat and prepare 3 to 4 smears. Stain with Wright stain.
- Examine each smear for the presence of LE cells under OIO. Report results as positive or negative.
