Lab 5 - Antimicrobials Flashcards
1
Q
What was the first instance of a microbe producing an antibiotic?
A
1897 Ernest Duchesne = Penicillum glaucum inhibited bacterial growth
2
Q
Which genus is responsible for the most antibiotics?
A
Streptomyces from Actinomycetes
3
Q
Explain how the cross-streak method works
A
- Obtain pure cultures of sample
- Grow isolate on agar
- Excreted antimicrobial diffuses thru agar
- Cross-streaked bacteria may be inhibited
4
Q
Is Streptomyces more effective against Gram negative or positive bacteria overall?
A
Gram negative (also effective against some Gram positive)
5
Q
Briefly describe the method of this experiment
A
- Inoculate TSA plates with 1 Streptomyces species each
- Incubate 10 days at room temperature
- Inoculate loopfuls of bacteria culture onto plates parallel to Streptomyces growth
- Incubate overnight at 37C
- Measure inhibition in mm and estimate growth density using given charts
6
Q
Which bacteria was consistently resistant to inhibition?
A
Pseudomonas aeruginosa
