What is the principle behind McMaster egg counting
It is a rapid/ simple quantitative method for counting parasite eggs in faeces, based on flotation on concentrated salt solution in a counting chamber
How do you perform McMaster egg counting
- Weigh 1g of faecal sample and add a known amount of flotation fluid, then mix by shaking
- Pour the faecal slurry through a sieve and flush a known amount of flotation fluid through the sieve
- Transfer this sample using a pipette to the 2 McMaster slide chambers taking care to avoid bubbles
- Leave the counting chambers to stand for 5 minutes before examination
Examine the filtrate in the filled chambers under the microscope at 4x followed by 10x magnification - Identify and count all eggs within the engraved area of both chambers
What is the calculation used in McMaster egg counting
Number of eggs/g faeces= (Number of eggs counted x (60) total volume))/ ((0.3) volume of counted chamber x (1) weight of faeces)
