Lab Tech Flashcards
1
Q
Direct Selection
A
A form of Mutant Selection where cells are inoculated into a selective agar medium that only promotes the growth of the mutatnt
2
Q
Indirect Selection
A
When the non-mutant is a prototroph but the mutant is a auxotroph, this method is used to isolate the mutant
3
Q
Steps for Replica Plating
A
- Sterilize velvet and press the master plate onto it
- inoculate a minimal agar medium and a nutrient agar medium
- observe the colonies that didn’t grow on the glucose-salt agar– those are the mutations
4
Q
Penicillin Enrichments
A
Selectively kills prototrophs before replica plating to increase the proportion of auxotrophic mutants
