Negative (E.coli)
Positive (E.aerogenes)

37
Q

what is the substrate for Citrate test?

A

Citrate: Ammonioum Phosphate

38
Q

what type of enzyme for Citrate test?

A

intracellular

39
Q

what enzyme is involved for Citrate test?

A

N/A

40
Q

What are the products for Citrate test?

A

NH3 or NH4

41
Q

what is a postive test for Citrate test?

A

Citrate permease allows cell to absorb citrate. Citrate is only C source thus allowing bacteria to grow. As they grow, they us (NH4)H2PO4 and pH increases. Green tube turns to blue.
Positive is blue ( E.coli)
negative is not blue ( E. aerogenes)

53
Q

For Catalase experiment what are we testing for?

A

Presence of the enzyme catalase

54
Q

What reaction does catalase catalyze?

A

converts hydrogen peroxide into water and gaseous oxygen

55
Q

which groups of organism posses catalase ?

A

B. subtilis

bacillus

56
Q

what does superoxide dismutase do?

A

catalyzes conversion of superoxide radicals to hydrogen peroxide

57
Q

why is oxygen toxic to strict/obligate anaerobes?

A

Strict anaerobic organisms die in the presence of oxygen because the presence of oxygen causes them to create superoxide. Superoxide is toxic and is dismantled in aerobes and facultative anaerobes by the presence of superoxide dismutase. However, strict anaerobes do not have this enzyme, leaving the cell with an accumulation of superoxide which will kill the organism.

58
Q

In the Nitrate Reduction experiment what enzyme we looking for?

A

Nitrate reductase

59
Q

what is Nitrate?

A

66
Q

If nitrate reduced completely what is it?

A
