Lab Quiz 2 Flashcards
What do Negative stains do?
Negative stain does not stain organism but stains the backgrounds.
how does a capsule appear in prepared slide?
clear surrounded by red stained background
Why is negative stained used?
to determin morphology and cellular arrangment in bacterica that are too delicate to withstand heat-fixing. (for example Treponema)
What color indicates bacteria is present in the agar? (Motility)
Red(TTC- Help us to visualize the bacterial in the agar)
What does the red(TTC) color repersent? (Motility)
bacterial metabolism
When oxidized what color is TTC?
colorless
When TTC is reduced? What does it mean?
it is red and forms an insoluble precipitate.
wherever you see red the bacterial are there.
What does TTC stand for ?
Tetrazolium chloride
What are wet mounts used for?
allow obersvation of living cells to determin motility. Also used to see natural cell size, arrangement, and shape
What is motility?
Move ones self
If it not motility, What is it?
Brownian movement (vibrating)
Two type of pigment for slants?
Intracellular( on top agar)
Extacellular (inside agar)
Two type of consistency for slants?
Mucoid (smooth and shiny)
Dry ( friable , crusty, and cracky)
Four type of growth patterns for broth?
Pelllicule (on top)
Tubridity (cloudy
sediment (at the bottom of the meduim tube)
Flaccutant(clump on side of tube or in agar)
What are growth patterns used for?
can provide useful information when attempting to identity an organism.
