Gram stain procedure and what Gram + and Gram - will look like
Apply material to slide, dry
Crystal violet 1 min, wash
Lugol’s iodine 1min, wash
Alcohol decolorizer 10-20 sec, wash
Safarin counterstain 10-45 sec, wash and dry
Gram + : purple, Gram -: pink
Blood agar
Common general isolation media, not selective
Differential for hemolytic action
Brain Heart Infusion agar
General purpose medium for growth of fastidious microorganisms, especially fungi and yeast
May see pigmentation
MacConkey agar
Selective for Gram - (but not all Gram - will grow)
