LAB 2 - The Microscope Flashcards
What are the 2 types of microscopes?
- Dissecting microscope (stereoscope)
2. Compound light microscope
What is a dissecting microscope used for?
-to view larger (or opaque) organisms
What is the magnification of a dissecting scope?
-low mag. (8x-35x)
Is a dissecting scope mono or binocular?
both
What is a compound light microscope used for?
-to magnify a very small object
What is the magnification of a compound light scope?
40x-1000x
Is a compound scope mono or binocular?
both
What is used to clean a microscope?
special lens cleaning paper only
What are the 4 magnification objects and their names?
- 4x scanning (40x)
- 10x low-power (100x)
- 40x high power (400x)
- 100x oil-immersion (1000x)
What mag. do you start with?
4x
What are the 2 types of slides?
- prepared (permanent)
2. wet-mount (temporary)
What is focusing a microscope?
- increasing/decreasing distance between specimen on slide
- increasing/decreasing objective lens
As magnification increases, the diameter of the field becomes ________
smaller
What is the equation for diameter of field of view?
D(a) x Mag(a) = D(b) x Mag(b)
What is the approximate Diameter of Field of View for each objective?
4x 4.0mm
10x 2.0mm
40x .4mm
100x .2mm
As magnification increases the depth of field _______
decreases
Which objective has the greatest depth of field?
scanning power
In lab, which slide was used to test depth of field?
different colored strings
What organism was used to practice focusing?
Daphnia (water flea)
What are the 3 major parts of a microscope?
- lens system
- adjustable light source
- focusing mechanism
What is the condenser?
focuses the light on the specimen
What is the iris diaphragm?
- built into condenser
- allows you to regulate light intensity
Wha is the light pathway?
- passes through sample, enters objective lens
- passes through o cular lens (eyepiece)
monocular vs. binocular
Monocular: singular ocular lens on rotating head
Binocular: 2 ocular lenses, moved together or apart
What is magnification?
indicates number of times the image of the sample is enlarged by microscope
What is resolution?
- ability to clearly distinguish objects which are close together
- expressed as distance (nm or micrometer)
What is blue light?
-utilized by microscopes because of short wavelength to increase resolution
Resolving power ______ as the wavelength of light used decreases
increases
What is working distance?
distance between the objective and the sample
What do parcentric and parfocal mean?
-when new lens is rotated in place, the image remains centered + close to focus
What is field of view?
diameter of visible image
What is depth of field?
distance between the highest and lowest points in the sample that are simultaneously in focus
What 3 things decrease with increasing mag.?
- depth of field
- field of view
- light intensity
