L8 Drug Elimination Flashcards
• Drugs are eliminated from the body either:
- Unchanged
or - Converted to metabolites
(By process of metabolism),
then the metabolites are excreted
Removal of a drug from the body may occur via a number of routes, the most important being the kidney or urine
• Other (less) routes:
- bile, intestine, lung, milk,
- Drugs that have been made water soluble in the liver are often readily excreted in the kidneys.
DRUG ELIMINATION/ EXCRETION
The time taken to reduce the plasma drug concentration by half
Elimination Half Life (T1/2)
the amount of drug cleared from the body per unit of time.
Drug Clearance (CL)
Drugs must be sufficiently polar to be eliminated from the body.
DRUG CLEARANCE BY THE KIDNEY
the most important organ for drug clearance is the kidney into the urine.
DRUG CLEARANCE BY THE KIDNEY
CLEARANCE BY OTHER ROUTES:
- the feces:Drugs that are not absorbed after oral administration or drugs that are secreted directly into the intestines or into bile.
- The lungs
- breast milk
- sweat, saliva, tears, hair, and skin
• the sum of all clearances
Total body clearance
