A

EdU is a thymidine homolog - incorperates into new DNA during synthesis (S-phase)
- it labels newborn cells

How can you lneage trace from a-tanycytes?

A
  • transgenic mouse - clone Cre-ERT2 (Cre fused to oestrogen ligand) downstream of a GLAST promoter
  • cross with a mouse which has a stop codon downstream of constitutive promoter (rosa26). and a GFP downstream of the STOP codon
  • when tamoxifen injected - cre-ERT2 is activated and cuts lox sites in GLAST expressing cells (a tanycytes)
  • cutting of lox allows GFP to be expressed
  • GFP is only expressed in cells expressing GLAST and their progeny (a-tanycytes and progeny)

A

yes

  • they can give rise to more tanycytic populations and can also give rise to neurons and astrocytes
  • they are neurogenic

how is neurogenesis under caged conditions in mice?

A

under normal caged conditions levels of neurogenesis are low

  • this because the mice are well looked after and in a rach environment - they are not stressed
  • stress promotes neurogenesis
