l8 Flashcards
1
Q
what makes a neural cell multipotent?
A
- if it can self renew
- can give rise to both neurons and glia
2
Q
what does EdU do?
A
EdU is a thymidine homolog - incorperates into new DNA during synthesis (S-phase)
- it labels newborn cells
3
Q
How can you lneage trace from a-tanycytes?
A
- transgenic mouse - clone Cre-ERT2 (Cre fused to oestrogen ligand) downstream of a GLAST promoter
- cross with a mouse which has a stop codon downstream of constitutive promoter (rosa26). and a GFP downstream of the STOP codon
- when tamoxifen injected - cre-ERT2 is activated and cuts lox sites in GLAST expressing cells (a tanycytes)
- cutting of lox allows GFP to be expressed
- GFP is only expressed in cells expressing GLAST and their progeny (a-tanycytes and progeny)
4
Q
can a-tanycytes self renew?
A
yes
- they can give rise to more tanycytic populations and can also give rise to neurons and astrocytes
- they are neurogenic
5
Q
how is neurogenesis under caged conditions in mice?
A
under normal caged conditions levels of neurogenesis are low
- this because the mice are well looked after and in a rach environment - they are not stressed
- stress promotes neurogenesis
