light microscopy
tissues illuminated by a beam of light, gives a larger view and reveals basic structure
electron microscopy
tissues illuminated by a beam of electrons , reveals cell ultrastructure but in a smaller view
what are the 7 steps to obtaining tissues for microscopy
1 - specimen collection 2 - fixation 3 - dehydration 4 - embedding 5 - sectioning 6 - staining 7 - viewing
punch biopsy
used to obtain specimen from skin or oral surfaces
needle biopsy
used to obtain specimens from organs or lumps below the skin
often used alongside X-ray/ MRI to guide biopsy
endoscopic biopsy
flexible tube inserted into incision of opening.
what is a common fixative
formaldehyde
why are samples dehydrated during tissue collection
most methods of embedding e.g paraffin are not compatible with water
microtome

haematoxylin and eosin

haem - stains acidic structures blue/ purple
eosin - stains basic structures pink/ red
