Identification of Yeasts and Molds Flashcards
IoY: Morphological includes:
a. color of colonies
b. size and shape of cells
c. presence of capsule around the cell
d. production of hyphae, pseudohyphae, arthroconidia, and(or) chlamydospores
Identification of yeast relies on a combination of
Morphological, Physiological and Biochemical characteristics
IoY: Biochemical characteristics
a. assimilation and fermentation of sugars
b. assimilation of nitrate
IoY: are based on sugar assimilation of isolates
Most commercial tests (eg., ID YST, bioMérieux)
IoY: (MALDI-TOF)2
Matrix-assisted laser desorption ionization-time of flight mass spectrometry
Most can be identified after growth in culture. Recognizable criteria for recognition often differ from the fundamental characteristics as the basic of classification.
1. Macroscopic characteristics viz. colonial form, surface color (obverse) pigmentation, reverse pigmentation, and growth rate.
Identification of Molds
IoM: Factors that influence the colonial form
- Culture medium
- Incubation temperature
- Age of culture
- Amount of inoculum
IoM: is important in speciation, hence it is important to select culture conditions which favor sporulation.
Sporulation
IoM: promotes over production of mycelium, which results in loss of sporulation, in such cases, subculture to a low-nutrient medium (stimulate sporulation)
Sabouraud’s dextrose agar (SDA)
