Experiment Overview/Pipetting Flashcards
1
Q
types of micropipettes?
A
- gilson (2 stops)
- eppendorf (3 stops)
1
Q
what are the steps of the experiment?
A
- Restriction digest of insert (LDL receptor)
- Transformation into E. coli JM101
- determine cholesterol conc in spent media via standard curve
- RD and plasmid map
2
Q
1st thing to do when setting pipette?
A
check range
3
Q
range and reading of each type of micropipette?
A
p2: 0.2µL-2µL (1 black digit, 2 red decimal places)
p20: 2-20µL (2 black digit, 1 red decimal place)
p200: 20-200µL (all black no decimal place)
p1000: 200/100-1000µL (highest is 100 = 1000, 20 = 200)
4
Q
if 2nd stop is used to draw liquid..
A
too much volume is drawn
5
Q
using pipette out of range causes…
A
- inaccurate volume measurements
- damage to internal pipette mechanisms
6
Q
using pipette without tip causes…
A
- contamination
- pipette corrosion
7
Q
What are the pipette tip types?
A
- yellow - P20 + P200
- blue - P1000
- filter tip - contains sterile filter within each tip to preent contamination
