Ex 2. L2: Multi Dose II Flashcards
1
Q
Dosing regimen steps
A
- Know the target concentration range
- Determine the max dosing interval (TauMax) round to reasonable interval (E.g., q6h, q8h, q12 and once daily)
- Determine maintenance dose (DM). Choose Cssave to fall in the middle of target concentration range
- Check whether Cssmax and Cssmin fall within the expected range. Adjust dosing regimen (dose or Tau) if they do not fall within the required range
- If necessary, determine loading dose (DL)
2
Q
1: Know the target concentration range
A
We know that at conc. above 100mg/L Drug X shows signs of toxicity. We also know that at conc. Less than 20mg/L, drug shows no effect.
Range must be between 20-100
3
Q
2: Determine the max dosing interval (Tmax), round to reasonable interval
A
Max time interval over which the upper and lower concentrations can occur
4
Q
- Determine Maintenance dose
A
(Dose/T)/CL * Tau * CL
5
Q
- Check whether Cssmax and Cssmin fall within expected range
A
Make sure it is good as the concentration fluctuates within the therapeutic range window
6
Q
- If necessary, determine loading dose
A
Time to reach SS
-Will reach ss after the second dose
-Loading dose is likely
unnecessary
7
Q
- If you do not want to wait to reach therapeutic concentrations: Loading dose
A
DM = DL - DL * e^-Kt
